摘 要:目的和方法:应用免疫荧光及免疫电镜技术,对慢性酒精投与之实验大鼠及重症酒精性肝炎患者肝脏枯否细胞对LPS的清除功能进行了观察。结果:经门静脉注射LPS10min后,Lieber’s酒精饮食投与6周之大鼠肝脏枯否细胞内抗LPS荧光显色低于正常大鼠,电镜下枯否细胞内抗LPS阳性颗粒少见,而肝窦壁内皮细胞及肝细胞内抗LPS阳性物增多。重症酒精性肝炎患者肝穿组织枯否细胞内抗LPS荧光显色亦显著低于正常对照。结论:酒精刺激后肝脏枯否细胞对LPS的清除功能降低。AIM and METHOD:The effects of alcohol on lipopolysaccharide (LPS) eliminating actions of liver Kupffer cells in rats with chronic alcohol-induced liver injury and in patients with severe acute alcoholic hepatitis were observed.RESULTS:Ten minutes after the injection of LPS(0.2 mg/kg body weight )via portal vein, Kupffer cells in the liver of normal rats showed strong FITC fluorescent reactions to LPS, but only slight increase of FITC fluorescent reactions to LPS was found in Kupffer cells in the liver tissues of chronic alcohol induced-liver injury rats. Under electron microscope, few positive reactions to LPS was found in Kupffer cells in the liver of alcohol injury rats, but positive reactions to LPS increased in sinusoidal epithelial cells and hepatocytes. FITC fluorescent intensity in Kupffer cells in the liver samples of patients with acute severe alcoholic hepatitis was significantly lower than of normal controls. CONCLUSION:Alcohol diminisk the LPS-eliminating actions of Kupffer cells in the liver, and LPS might play a role in the mechanism of liver injury induced by alcohol.
