摘  要:目的观察白藜三醇(Res)对人外周血内皮祖细胞(EPCs)功能的影响。方法密度梯度离心法获取人外周血单个核细胞,培养7d后,收集贴壁细胞并加入Res(1、10、25、50μmol/L)干预一定时间(6、12、24、48h)。激光共聚焦显微镜和流式细胞术鉴定EPCs,分别观察EPCs的增殖、黏附、迁移和体外血管生成能力。结果Res促进外周血EPCs扩增,50μmol/LRes作用24h对EPCs数量的影响最为显著(P<0.01)。Res也显著改善了外周血EPCs的增殖、黏附、迁移和体外血管生成能力。结论Res可增加EPCs数量并改善其功能。Objective To investigate the effects of resveratrol on the activity of endothelial progenitor cells(EPCs) from human peripheral blood. Methods EPCs were isolated from human peripheral blood and identified immunoeytochemically. EPCs were incubated with resveratrol (1,10,25 and 50 mmol/L) or placebo for 7 days. Cell proliferation, migration and in vitro vasculogenesis were observed using MTT assay, modified Boyden chamber assay and in vitro vasculogenesis detection, respectively. Results Incubation of isolated human MNCs with resveratrol increased the number of EPCs, maximum at 50μmol/L, 24 h(approximately 1-fold increase (P〈0. 01). In addition, resveratrol promoted the capacity of EPCs proliferation, adhesion, migration and in vitro vasculogenesis. Conclusion Resveratrol may increase the number of EPCs and improve the functional activity.
