摘 要:目的探讨银杏叶提取物(EGB)对大鼠局灶性脑缺血/再灌注中脑组织内血管内皮细胞生长因子(VEGF)及血管通透因子(HIF-1)表达的影响。方法采用Zea Longa线栓法制备大鼠局灶性脑缺血/再灌注模型(即MCAO模型),54只大鼠随机分为三组,即对照组(n=6)、EGB组(n=24)及缺血/再灌注组(n=24),分别在缺血/再灌注4、6、24、72h取材。应用免疫组化法(SABC法),利用计算机图像分析系统软件,计数脑梗死后不同时间点阳性细胞的数量。检测VEGF及HIF—1蛋白的表达变化情况。结果对照组:脑组织VEGF和HIF表达均较低;EGB组:VEGF及HIF阳性细胞密度均相对较高;缺血/再灌注组:VEGF在4h开始升高,24h达到高峰,72h下降到接近正常水平,HIF-1在缺血后4h开始降低,8h开始升高,24h达到高峰,72h下降到接近正常水平。结论EGB对于缺血性脑损伤有保护作用。Objective To observe the effect of extract of Ginkgo biloba ( EGB ) on the expression of vascular endothelial growth factor ( VEGF ) and HIF - 1 after focal cerebral ischemia reperfusion in rates. Methods MCAO ( Middle Cerebral Artery Occlusion ) models were set up in rats. 54 male Wistar rates were randomized into three groups : control group, EGB groups and groups of isehemia for 2 h then respective reperfusion for 4 h, 6 h, 24 h, 72 h. tmmunohistochemistry (SABC)were used to detect the VEGF and HIF - 1 protein expression. Results VEGF and HIF - 1 protein expression in control group were remarkably deep, and it was strongest in EGB groups. VEGF increased at 4 h, reached peak at 24 h, then returned to normal level at 72 h, while HIF - 1 increased at 8 h , reached peak at 24 h, then returned to normal level at 72 h. Conclusion EGB has neuroprotective effect against cerebral ischemie injury.
