摘 要:目的观察丙氨瑞林对子宫内膜癌裸鼠移植瘤生长的抑制作用及机制。方法体外培养人子宫内膜癌Hec-1-B细胞。将Hec-1-B细胞接种于左下肢近腹股沟处皮下,建立子宫内膜癌动物模型。将24只荷瘤裸鼠随机分为4组,即低剂量组(13μg/kg),中剂量组(26μg/kg),高剂量组(52μg/kg)和空白对照组(0μg/kg),每组6只,造模3d后开始用丙氨瑞林干预,给药途径为皮下干预,每天1次,持续4周。治疗期间定期测量肿瘤大小,观察裸鼠全身状况,实验结束时,将移植瘤完整取出,称取瘤重,测量肿瘤体积、绘制肿瘤生长曲线、计算抑瘤率,并用反转录-聚合酶链反应(RT-PCR)法检测PtenmRNA的表达。结果①当丙氨瑞林浓度为13μg/kg时,癌细胞即受到抑制,抑瘤率为32.4840%;随着丙氨瑞林浓度的增大,抑制率渐高。到浓度为52μg/kg时抑瘤率上升至35.4494%,与对照组比较差异有统计学意义(P<0.05)。②浓度从13~52μg/kg的丙氨瑞林作用4周后,Pten的表达有不同程度的上升,与对照组相比,差异有统计学意义(P<0.05)。结论丙氨瑞林对裸鼠移植瘤生长可能有直接的抑制作用,且呈剂量依赖关系。丙氨瑞林抑制作用的分子机制可能与上调Pten蛋白表达有关。Objective To study if Alarelin can inhibit nude mice bearing human endometrial carcinoma and to explore its molecular mechanisms. Methods Human endometrial carcinoma hec-1-b cells were cultured in vitra. They were inoculated to 24 athymic mice at the left lower extremity near the fold inguen. Than the mice were randomly divided into blank control group, low-dose group, middle-dose group and high dose group. After 3 days of the inoculation, we injected subcutaneouly the mice of low-dose group( 13 ~t g/kg ), middle-dose group(26 ~t g/kg) and high dose group(52 μ g/ kg) with Alarelin one time every day for 4 weeks and the blank control group were injected with the isodose isotonic Na chloride in the same way. During the period of injection with Alarelin, we measured the tumor size and observed the condition of the mice. At the end of the experimentation, the tumors were dislodged completely then we measured the weight of tumors, drawed the growth curve of the tumors, calculated the inhibition rate of the tumor and detected the expression the PTEN with the method of RToPCR. Results 0)The growth of tumor was inhibited when the lowest Alarelin concentration 13 ta g/kg was given, inhibition rate of the tumor was 32.4840%. The inhibition rate of the tumor increased with the rising of Alarelin concentrations. When Alarelin concentration was 52 μg/kg, the inhibitory rate increased to 35.4494%, the difference was significant (P〈0.05). After 4w with 13 μ g/kg to 52 μg/kg Alarelin, the level of pten increased. The difference was significant(P〈0.05). Conclusion ①Alarelin might have directed inhibitory response on nude mice bearing human endometrial carcinoma and the effect was dose-dependent. ②Up-regulation of Pten protein may be one of the mechanisms through which Alarelin induced growth inhibition of nude mice bearing human endometrial carcinoma.
