双重组合染色法对兔脊神经节细胞尼氏体形状的染色观察

摘  要:目的:调整染色技术观察兔脊神经节细胞尼氏体常态下的形态与结构特征。方法:通过煌焦油紫(Bright cresyl violet)、甲苯胺蓝(Toluidine blue)、硫堇(Thionin)和橙黄G、磷钨酸(Orange G,Phosphotungstic acid)双重组合染色法(简称BTT-OP法)显示脊神经节(C1~L7)细胞尼氏体形状。结果:小细胞尼氏体染色多呈均匀细颗粒状;大、中型细胞的尼氏体形状分3种:(1)尼氏体呈小块状围绕核呈同心圆状排列;(2)尼氏体呈小块状,在核周均匀分布;(3)尼氏体呈小颗粒状。结论:BTT-OP染色法可清楚显示兔脊神经节细胞尼氏体形态结构,通过观察发现脊神经节大、中、小细胞尼氏体形状与排列不同。Objective: To observe the Nissl bodies shape and structure in rabbit spinal ganglion (C1-L7). Methods: The double-compound staining dyes composed of brilliant cresyl violet, toluidine blue, thionin, Orange G and phosphotungstic acid (BTT-OP) were used. Results:The Nissl body of the small neuron presented fine-grain shape. The large and medium neuron presented three kinds: the Nissl body presented the block shape and were concentric circles around the nuclear. The Nissl body presented the block shape and were uniform distribution around the nuclear, the Nissl body presented fine-grain shape. Conclusion: It shows clearly the Nissl bodies shape and structure in rabbit spinal ganglion by BTT-OP methods. This result provides the shape of the Nissl body of the small neuron,the large and the medium neuron are different.
