摘  要:阿里红多糖具有提高机体免疫能力、抗衰老等作用,广泛用于医药、保健品、食品添加剂等领域。研究利用新疆稀有药用拟层孔菌阿里红(Fomes officinalis Ames)野生菌株进行液体深层发酵生产胞外多糖,通过单因素及正交实验对种子和摇瓶发酵培养基的营养成分、配比浓度、生长因子及各因素的最佳组合等进行了筛选优化。根据菌丝体生长和胞外多糖产量情况,最终确定种子培养基:葡萄糖3%、蛋白胨1.5%、KH_2PO_40.3%、MgSO_4·7H_2O 0.1%、VB_1 0.1 mg/mL;发酵培养基:葡萄糖1%、麦芽浸粉0.3%、蛋白胨0.75%、酵母浸粉0.5%。经12 d发酵,每50 mL发酵液菌体干重和胞外粗多糖含量最高可达65.4和77.7 mg。Extracellular polysaccharide of Fomes Officinalis Ames were used widely with the functions of immune- enhancing and anti- senescence. This study was conducted to optimize the nutrient component, mixture ratio and concentration, growth factor and best combination of all elements through single factor and orthogonal test to produce extracellular polysaccharide by the wild strain of Fomes offcinalis Ames of Uygur Traditional Medicine. The optimal content of seed and fermentation media were obtained according to the yield of biomass and extracellular polysaccharide were as follows, seed medium: glucose 3%, peptone 1.5%, KH2PO4 0.3%, MgSO4·7H2O 0.1%, VB1 0.1 mg/mL; fermentation medium: glucose 1%, malt extract 0.3%, peptone 0.75%. The results showed that with the optimized seed and fermentation media, the content of dry weight and extracellular polysaceharide reach 65.4 mg and 77.7 mg per 50 mL broth content, respectively when it was fermented for 12 d.
