摘  要:目的了解近年引起食物中毒的26株副溶性弧菌分离株的携带主要毒力基因的分子流行病学特征和耐药性状况,为制定相应的防控措施提供参考。方法根据副溶血性弧菌耐热直接溶血素(TDH)和耐热相关溶血素(TRH)基因序列设计PCR引物,建立PCR反应体系,检测26株从2005年11月至2008年8月发生的8起散发性食物中毒事件中分离的副溶血性弧菌菌株的毒力基因TDH和TRH,K-B法测定其药物敏感性。结果26株副溶血性弧菌有23株携带有TDH基因,阳性率为88%,所有菌株均未带有TRH基因;26株菌对亚胺培南、阿米卡星、头孢吡肟、头孢噻肟、庆大霉素、环丙沙星、哌拉西林、头孢哌酮、头孢他啶、头孢西丁等13种药物未见有耐药菌株;但对磺胺异恶唑、替卡西林、氨苄西林等有较高的耐药率,分别达76.9%、69.2%、80.8%。而对复方新诺明、四环素、头孢呋辛酯、头孢噻吩、头孢唑啉等也有耐药菌株的存在。结论带有TDH基因是近年引起食物中毒的副溶性弧菌分离株的主要分子流行病学特征,所分离菌株对多种常用药物敏感性高。研究结果为评价引起食物中毒的副溶性弧菌的分子流行病学特征和制定可行的防控措施提供了科学依据。Objective To study the molecular epidemiology of major toxin-genes and the drug-resistance of 26 strains of Vibrio parahaemolyticus isolated from 8 foodborne-disease incidents in Shenzhen since Nov 2005 to Aug 2008. Method PCR primers were designed and PCR methods were established to examine the genes of thermostable direct hemolysin (TDH) and TDH-related hemolysin (TRH). Drug resistance was tested by using Kelly-Braiy method. Result 88% (23/26) of Vibrio parahaemolyticus isolates were positive for the presence of TDH gene and all isolates were negative for the TRH gene. Results showed that these isolated strains were highly sensitive to 13 antibiotics such as imipcnem (100%), Amikacin (100%), cefepime (100%), cefotaxime (100%), Gentamiein (100%), ciprofloxacin (100%), Piperacillin (100%), cefoperazone (100%), cefiazidime (100%) and cefoxitin (100%). Susceptibility of these isolates to some frequently used medicines, such as sulfafurazole (SF), Ticarcillin (TIC) and ampicillin (AMP), was low. The percentage of drug-resistant strains to these three drugs was 76.9%, 69.2% and 80.8%, respectively. Drug-resistance to co-trimoxazole, tetracycline, cefuroxime axetil, cephalothin and cefazolin was also found. Conclusion These results suggest that most of the bacteria isolated from foodborne-disease incidents were positive for the TDH gene and were susceptible to several frequently-used antibiotics. These results would provide foundation for the evaluation of molecular epidemiological characteristics of foodbome Vibrio parahae mol yticus .
