摘 要:目的观察GSK-3β功能改变对胃癌细胞生长影响并初步探讨其作用机制。方法体外MTT观察LY294002单独或联合LiCl对人胃癌细胞株SGC-7901作用。采用Western blot、RT-PCR和免疫细胞化学法分别检测总GSK-3β、p-GSK-3βser9、CyclinD1和Caspase3表达。结果LY294002对SGC-7901细胞增殖具有抑制作用,呈时间浓度依赖关系;LY294002干预细胞72h后,总GSK-3β表达无改变,p-GSK-3βser9和CyclinD1表达下降,caspase3表达升高;LiCl部分拮抗LY294002对细胞的生长抑制及对CyclinD1和caspase3表达的改变。结论GSK-3β对胃癌细胞生长具有抑制作用,其机制可能与阻滞细胞周期和诱导细胞凋亡有关。Objective To observe the effect of GSK-3βon the proliferation of human gastric carcinoma cell and its mechanism. Methods Cell proliferation of human gastric carcinoma cell line SGC-7901 treated by LY294002 with or without LiCl was analyzed by MTT assay in vitro. The expression of total GSK-3β, p-GSK-3β^ser9, CyclinD1 and Caspase3 was detected by western blotting, RT-PCR and SP immunohistochemistry respectively. Results LY294002 could inhibit the cell proliferation dependent with concentration and time;the expression of total GSK-3β was constant, the protein level of p-GSK-3β^ser9, the CyclinDlmRNA and protein level were down-regulated and the expression of caspase3 was increased after treated SGC-7901 with LY294002 for 72h. The growth inhibition and effect on the expression of CyclinD1 and Caspase3 by the LY294002 were partially abrogated by lithium chloride. Conclusion GSK-3β could negatively regulate the proliferation of human gastric carcinoma cell. The possible mechanism is that GSK-3β could lead to cycle arrest and induce apoptosis.
