Line 10肝癌细胞产生的肿瘤生长因子对人肝上皮癌细胞体外增殖的影响

摘　　要：目的：检测近交２系豚鼠肝癌细胞ｌｉｎｅ１０分泌的生长因子ＴｕＧＦ（ｓ）对人肝上皮癌细胞（ＢＥＬ－７４０２）生长的影响。方法：集落形成测定法检测ＴｕＧＦ（ｓ）对ＢＥＬ－７４０２细胞在软琼脂培养基内生长的影响。ＭＴＴ法测定ＴｕＧＦ（ｓ）对ＢＥＬ－７４０２细胞在液体培养基内增殖的影响。结果：软琼脂培养基内培养时ＴｕＧＦ（ｓ）对ＢＥＬ－７４０２细胞无集落形成促进或抑制作用；单层培养的条件下，ＴｕＧＦ（ｓ）对ＢＥＬ－７４０２细胞在１×１０５个／ｍｌ接种细胞浓度下，具有促进细胞生长的作用；而在１×１０４个／ｍｌ细胞浓度下，则为抑制细胞生长的作用。Objective:Line 10 hepatoma cells of strain 2 guinea pig can secret growth factors(TuGF(s)). The effects of TuGF(s) on the growth of human liver epithelioma cells (BEL7402) were tested. Methods:The effects of TuGF(s) on the growth of BEL7402 cells in the soft agar medium were tested by colonyforming assay. The activities of TuGF(s) on the growth of BEL7402 cells in the monolayer culture were estimated using MTT methods. Results:TuGF(s) showed little effect on BEL7402 cell lines in the soft agar culture system. TuGF(s) promoted the growth of BEL7402 cell lines in the monolayer culture in 1×105 cells/ml concentration while inhibited it in 1×104 cells/ml. Conclusions:[WT5”BZ〗TuGF(s) have sophisticated biological regulatory effects on the growth of BEL7402 cell lines depending on the experiment conditions.
