摘 要:目的:研究机械张应变对人牙髓细胞L型钙离子通道基因表达的影响。方法:取新鲜拔除的健康恒牙牙髓,采用组织块贴壁法培养原代人牙髓细胞,通过Flexcell细胞应力培养系统,分别施加2%和8%的周期性张应变,加载时间分别为0.5、12、24 h,观察机械牵张力对人牙髓细胞L型钙离子通道不同亚型表达的影响。结果:人牙髓细胞(HDPCs)主要表达L型钙离子通道α1亚基C亚型(L type calcium channelα1 Csubunit,Cav1.2)和L型钙离子通道α1亚基D亚型(Ltype calcium channelα1 D subunit,Cav1.3),未见L型钙离子通道α1亚基S亚型(L type calcium channelα1 S subunit,Cav1.1)和L型钙离子通道α1亚基F亚型(Ltype calcium channelα1 F subunit,Cav1.4)的表达。在2%和8%张应变下,加载0.5 h至12 h Cav1.2和Cav1.3均有上调(P<0.05),而加载24 h后2个亚型与对照组间均无明显差异(P>0.05)。结论:人牙髓细胞主要表达Cav1.2和Cav1.3,应力和作用时间对L型钙离子通道表达量有明显影响。AIM : To investigate the effects of mechanical stress on expression of L - type calcium channel in human dental pulp cells in vitro. METHODS: Human dental pulp cells were cultured and subjected to 2% or 8% cyclical strain with Flexcell Tension Plus System for 0.5h, 12h and 24h respectively, and then the expression of L type calcium channel were examined. RESULTS: L type calcium channel α1 S subunit and F subunit were not detected under the tested condition, while L type calcium channel α1 C subunit and D subunit were expressed in HDPCs, and upregulated by 2% and 8% strain for 0.5h and 12h( P 〈 0.05 ). CONCLUSION: Human dental pulp cells mainly express Cavl. 2 and Cav1. 3 subtypes of L- type calcium channel ,which could be regulated by mechanical stress in a time - dependent manner.
