芦荟液-表皮生长因子复合医用胶促进大鼠口腔溃疡愈合的研究

摘　　要：目的：研究表皮生长因子（Epidermal growth factor，EGF）和芦荟333对细胞生长的影响，并观察基于医用胶的芦荟液-EGF复合药膜促进实验性口腔黏膜溃疡愈合的作用，为制备新型口腔复合膜提供实验依据。方法：用MTT法分别检测不同浓度芦荟和（或）表皮生长因子在体外对3T3细胞增殖的影响，并确定最佳应用浓度。之后建立大鼠的口腔黏膜溃疡模型，观察医用胶联合EGF和芦荟液对黏膜溃疡的治疗效果。结果：①EGF在0．1～10ug／L浓度范围对3T3细胞作用24h呈抑制生长作用；48h和72h时EGF对细胞作用无显著差异。②芦荟液汁对333细胞生长有显著的促进作用。选择芦荟液汁1：100稀释，作为动物实验的给药剂量。③10ug/LEGF对芦荟促进3T3细胞生长有协同作用。④EGF＋芦荟、EGF＋芦荟和医用胶组用于小鼠口腔黏膜溃疡治疗效果显著，同时EGF＋芦荟和医用胶组具有给药次数少，愈合效果显著的特点。结论：EGF、芦荟和医用胶复合药膜可明显促进实验性口腔黏膜溃疡愈合，具有一定的临床实用性。AIM：To evaluate the biological effects of epidermal growth factor （EGF） and aloes on cell prolifera- tion, and then to develop medical adhesive based compound oral pellicles to treat oral ulcer and observe its therapeutic effect. METHODS：Thiazoly blue （MTT） colorimetric assay was used at different time points（24, 48 and 72h） to determine the proliferative effect after administration of EGF and aloes with different concentrations. Then oral ulcer was established in SD rats by local administration of crystal NaOH. Totally 90 rats were divided into 6 groups with 15 in each. The oral ulcer in each group was treated by saline, EGF, aoles or medical adhesive along or combination . After 3 - and 7 - days treatment the therapeutic effect was observed and histopathological changes were checked by HE staining. RESULTS： （1）In comparison to the control group ,0. 1 - 10ug/1 EGF significantly inhibited the proliferation of 3T3 cells at 24h. （2）During 24 - 72h compared with control group ,aoles（1：100） significantly stimulated the proliferation of 3T3. （3）10ug/l EGF and aoles have important roles of cooperation or to adjust. （4）The treatment groups of EGF ＋ aoles and EGF ＋ aoles ＋ medical adhesive ef- fectively accelerated the healing time of oral ulcer. The healing rate of the EGF ＋ aoles ＋ medical adhesive group was also higher than other groups with only single administration. CONCLUSION： The compound oral pellicles by EGF, aloe and medical adhesive could promote healing of oral ulcer.
