摘  要:目的观察针刺对大鼠局灶性脑缺血再灌注损伤后海马神经元内细胞周期蛋白依赖性蛋白激酶4(CDK4)和神经元凋亡的影响。方法大脑中动脉线栓法建立脑缺血再灌注损伤模型,再灌后针刺大鼠,应用免疫组化、免疫印迹法和流式细胞计数法分别检测CDK4和细胞凋亡。结果缺血组再灌注48h后海马CDK4表达升高,凋亡细胞增多,针刺组CDK4表达和凋亡细胞明显减少(P<0.01)。结论针刺对脑缺血再灌注损伤有保护作用,其机制可能与调控细胞周期因子从而抑制凋亡有关。Objective To investigate the effects of acupuncture on expression of CDK4 protein in neu,, rons and neuronal apoptosis after focal cerebral ischemia reperfusion in rats. Methods A model of middle cerebral artery occlusion (MCAO) in rats was established with intraluminal filament occlusion. Acupuncture was conducted on rats after reperfusion. The immunohistochemistry, western blot and flow cytometry were used respectively to detect the expression of CDK4 protein and neuronal apoptosis in hippocampus. Results A signifi- cant increase in CDK4 immunostaining and apoptotic neurons was detected at 48h after reperfusion. The differences of immunoreactivity of CDK4 and apoptotic neurons between ischemic group and acupuncture group were significant( P 〈 0.01 ). Conclusion Acupuncture can protect cerebral tissues by regulating cell cycle factors and inhibiting cell apoptosis in ischemia reperfusin injury.
