摘　　要：目的研究TSC2基因的表达产物tuberin在结直肠癌中mRNA和蛋白水平的表达及其与临床病理特征的关系,探讨其在结直肠癌发生中的作用。方法采用实时荧光定量逆转录聚合酶链反应方法检测40例结直肠癌与癌旁正常肠黏膜组织中tuberin mRNA的表达,应用免疫组织化学方法检测60例结直肠癌与癌旁正常肠黏膜组织tuberin蛋白的表达,分析tuberin mRNA和蛋白表达与临床病理特征的关系。结果40例结直肠癌中tuberin mRNA表达量与患者年龄、性别、肿瘤大小、组织学分级、临床分期、淋巴结转移、远处转移无相关性(P>0.05),但显著低于癌旁正常组织,差异有高度统计学意义(P﹤0.01)。tuberin蛋白在60例结直肠癌与癌旁正常肠黏膜组织中的阳性表达率分别为41.6%、75%,差异有统计学意义(P﹤0.05)。tuberin蛋白阳性表达率与性别、组织学分级相关。结论tuberin在结直肠癌中表达下调,可能参与结直肠癌的发生。Objective To investigate the expression of tuberous sclerosis complex 2 gene product tuberin, in colorectal cancer and explore the relationship between expression of tuberin and the pathogenesis of colorectal cancer. Methods Real-time quantitative reverse transcnptase-polymerase chain reaction was applied on 40 cases of colorectal cancer, 40 cases of corresponding adjacent noncancerous tissues in order to detect mRNA expression of tuberin gene, protein expression was detected by immunohistochemistry. Statistical analysis was carried out to clarify the correlation between tnberin gene expression and various clinical parameters in these colorectal cancer patients, Results Tuberin mRNA in 40 colorectal cancer tissues was lower than that in all corresponding adjacent noncancerous tissues（P〈0.01）, there was a decreased tuberin protein expression in colorectal cancer. A significant difference of tuberln protein expression was also observed in colorectal cancer and corresponding adjacent noncancerous tissues （P 〈 0.05）, gender and clinical stage were correlated significantly with tuberin protein expression. Conclusion Decreased expression of tuberin gene may play an important role in the carcinogenesis of colorectal cancer.
