摘  要:以剑麻株芽或幼叶为外植体,研究了不同培养基、不同培养温度和光照条件下,剑麻愈伤组织的诱导和生长特性.结果证明,N6培养基诱导剑麻愈伤组织启动最快;MS、LS培养基无论对愈伤组织的诱导还是继代培养都具有良好的效果;B5培养基不适于剑麻组织培养.剑麻愈伤组织诱导和生长最适宜的温度为(25±2)℃、光照为12h.剑麻愈伤组织细胞可诱导产生蛋白酶.By choosing bulbils and seedling leaves of Agave sisalana as explants, different kinds of media and culture temperatures and lights for callus inducing and growing were studied. The results showed that among the four kinds of media, N 6 medium was the earliest one in inducing callus. MS medium and LS medium were suitable for callus inducing and growing. B 5 medium was unsuitable for tissue culture of Agave sisalana. For callus inducing and growing, the optimum temperatures were about (25±2)℃ and optimum illuminance was 12 h light. the callus could produce agavain_SH.
