摘 要:确定了用复合酶水解鱼鳞胶原蛋白的最佳条件和采用膜技术制备胶原蛋白低聚肽的工艺流程。对影响酶解的主要因素(酶种类、酶解时间、酶解温度、酶用量)进行了研究,通过正交实验确定最佳条件为:采用中性蛋白酶和木瓜蛋白酶组成的复合酶,酶解温度为60℃,酶解时间为4h,酶用量为2%。酶解液采用三级膜纯化和分离,再经冷冻干燥,最终得到纯度≥95%的胶原蛋白低聚肽,其中分子量500-800Da的肽的含量超过80%,产品质量高于国家标准。The optimum hydrolysis conditions for collagen and the membrane technological process for preparation of oligo-peptide of collagen were studied. The optimum hydrolysis conditions obtained by orthogonal experiment were as follows:using the compound enzyme made by proteolytic enzyme and papain enzyme, the enzymatic hydrolysis temperature was 60℃ ,the enzymatic hydrohysis time was 4 h,the enzyme amount was 2%. The enzymolyzed liquor was purifed and separated through three-stage membrane process, and oligo-peptide of collagen with purity more than 95 % was obtained after freezing and drying,in which the content of peptide with molecular weight of 500-800 Da was over 80% ,higher than the national standard.
