摘 要:FD-TRT是一种耐热逆转录酶(是自行表达并提纯的),根据TapDNA聚合酶的晶体结构为模板,采用计算机同源模建的方法建立FD-TRT的三级结构模型,并对FD-TRT和DNA聚合酶家族的其他蛋白功能差异的结构基础以及保守性功能性残基的作用进行研究,从而为进一步改造FD-TRT分子、提高其耐热性、反应活性以及对RNA模板和DNA模板的选择性提供良好的基础.In this paper, by comparing three automatic homology modeling medhods and taking the crystal structure of Taq polymerase as model block, the authors adopt a combined method to build a tertiary structure model of FD-thermostable reverse transcriptase (FDTRT), which was constructed and expressed by their laboratory earlier. The model makes them possible to investigate the structure basis for the functional difference between FDTRT and other proteins in DNA polymerase family. Furthermore, they also study the role of the reserved functional residue on the tertiary structure level. Thus some clues have been revealed for the further improvement of FD-TRT, such as raising its thermostability and reaction activity, offering good basis for the selectivity of RNA model block and DNA model block.
