摘　　要：目的研究喉癌患者外周血CK19mRNA、CK20 mRNA的表达,探讨其临床意义。方法采用巢式RT-PCR技术检测喉癌患者术前外周血中CK19mRNA、CK20 mRNA的表达。结果48例喉癌患者外周血中CK19 mRNA阳性率为37.5%(18/48),CK20 mRNA阳性率45.8%(22/48)。10例肿瘤组织中CK19 mRNA阳性率为100%(10/10),CK20 mRNA阳性率80%(8/10);10例声带息肉患者外周血为阴性对照,CK19 mRNA阳性率为10%(1/10),CK20mRNA阳性率为0。结合临床病理资料,喉癌患者外周血CK19 mRNA、CK20 mRNA表达与喉癌T分期、临床分期及淋巴结转移存在相关性,差异有统计学意义(P<0.05)。结论采用巢式RT-PCR技术,对喉癌患者外周血单个核细胞CK19mRNA和/或CK20 mRNA的检测,在分子生物学水平对喉癌细胞对周围组织的浸润及局部扩散提供了参考,有助于指导临床治疗。Objective To investigate the expressions of CK19mRNA and CK20 mRNA in peripheral blood of patients with laryngeal squamous cell carcinoma and their clinic significance. Methods The nested reverse transcription polymerase chain reaction was used to detect the expressions of CK19mRNA and CK20 mRNA in 50 cases laryngeal squamous cell carcinoma. Results The positive expressions of CK19mRNA and CK20 mRNA in peripheral blood of 50 cases were 37.5%（18/48） and 45.8% （22/48）. The positive expressions of CK19mRNA and CK20 mRNA in tumor of 10 cases of positive control were 100%（10/10） and 80%（8/10） and the results were 10%（1/10） and 0 （0/10） in negative control.The correlations between the expressions of CK19mRNA and CK20 mRNA in peripheral blood of patients with laryngeal squamous cell carcinoma and the T stage, clinic stage and metastes of local lymph node were significant （P〈0.05）. Conclusion The detection of CK19mRNA and/or CK20 mRNA in peripheral blood of patients with laryngeal squamous cell carcinoma by nested RT-PCR is valuable in guidance of clinical treatment.
