摘 要:目的观察花椒多酚类化合物总提取物(ZPPC)对活性氧自由基.OH、O2-.和肝脂质过氧化损伤的作用。方法分别利用化学发光法、体外肝匀浆温育Fe2+和H2O2诱导及CC l4肝损伤实验探讨ZPPC对.OH和O2-.的清除作用及检测肝和血浆中超氧化物歧化酶(SOD)活性以及脂质过氧化物丙二醛(MDA)含量。结果ZPPC能有效地清除活性氧自由基;口服低剂量ZPPC能显著降低CC l4中毒小鼠血浆MDA,提高SOD活性。结论适量ZPPC具有抗活性氧自由基和脂质过氧化的作用。Objective To study the effects of procyanidins extract from Zanthoxylum bungeanum Maxim. (ZPPC) on active oxygen radicals and lipid peroxidation. Methods (1) The scavenging effects of ZPPC on . OH, and O2^- were investigated by chemiluminescence method; (2) The contents of malondialdehyde (MDA) in liver homogenates incubated with Fe^2+ and H2O2 in vitro was determined; (3) The mice were treated with a single intragastric feeding of 5% CCl4 soya bean oil solntion (10 ml/kgbw) after seven days supplementation of ZPPC [200,400,800 mg/(kg bw. d)]. The activity of superoxide dismntase (SOD) and the content of MDA in liver and plasma of CC14 toxic mice were determined. Results In vitro, ZPPC 388.9 mg/L and 431.22 mg/L has marked activity on . OH in Fenton systems and O2^-. produced in xanthine/xanthine oxidase respectively, and could significantly decrease the contents of liver MDA. In vivo, ZPPC [ 200,400 mg/(kg bw . d) ] evidently reduced the content of MDA and increased the activity of SOD. Conclusion It suggests that ZPPC may be used as an antioxidant and might effectively protect liver from the injury due to lipid peroxidation.
