消瘀化痰方对非酒精性脂肪肝大鼠肝细胞凋亡和Caspase3蛋白表达的影响

摘　　要：目的观察中药消瘀化痰方对非酒精性脂肪肝(NAFLD)大鼠肝细胞凋亡和肝组织caspase 3蛋白表达的影响,探讨其防治NAFLD的部分作用机理。方法采用高脂饮食喂饲大鼠复制NAFLD模型,以东宝肝泰为对照药,运用流式细胞仪观察各组大鼠肝细胞凋亡及肝组织caspase 3蛋白表达的变化,同时采用HE染色观察各组大鼠肝组织形态学变化。结果镜下可见模型组大鼠肝脏中度以上脂肪变性,并有少量的肝细胞坏死;流式细胞术分析可见其细胞凋亡率明显增高(P<0.01),caspase 3蛋白表达FI值增高(P<0.01)。与模型组比较,各用药组肝脂变程度显著改善,肝细胞的凋亡率明显下降(P<0.01),caspase 3蛋白表达FI值降低(P<0.01)。结论消瘀化痰方通过抑制caspase 3蛋白的表达,抑制肝细胞的凋亡,以达到抗脂肪肝作用。Objective To observe the effect of Xiao - Yu - Hua - Tan - Yin on hepatic apoptosis and caspase3 in nonalcoholic fatty liver rats （NAFLD） . Methods The nonalcoholic fatty liver rat model was fed by high fat forage, and Dongbaogantai was treated as control group. Flow cytometry （FCM） was used to detect the hepatic apoptosis and expression of caspase3, and optical microscope was used to observe the degree of hepatic steatosis. Results Liver cells showed medium or serious steatosis by optical microscope, and a few hepatic necrosis. The rate of cell apoptosis and expression of caspase3 detected by FCM in the model group was significantly increased（ P 〈 0.01 ）. Compared with the model group, liver fat cell denaturizing was improved, and the rate of cell apoptosis and expression of caspase3 was significantly decreased in each treatment group（P 〈 0.01 ）. Conclusion Xiao - Yu - Hua - Tan - Fang can inhibit hepatic apoptosis inhibiting by the expression of caspase3, and protect nonalcoholic fatty liver.
