摘  要:目的探讨急性脑梗死(ACI)患者血浆谷胱甘肽过氧化酶(GPx)浓度的改变及其与GPx-3启动子基因多态性的关系。方法检测94例ACI患者和80例正常对照者血浆GPx浓度。在ACI患者发病第1d进行神经功能缺损程度评分(NDS)。采用聚合酶链反应-连接酶检测方法(PCR-LDR)检测ACI患者GPx-3启动子基因多态性。结果ACI组发病第1d血浆GPx浓度明显低于正常对照组(P<0.01),发病第14d时血浆GPx浓度与正常对照组比较,差异无统计学意义(P>0.05)。有GPx-3启动子基因多态性的ACI患者,血浆GPx浓度和NDS明显低于无GPx-3启动子基因多态性的患者(P<0.05~0.01)。结论ACI患者血浆GPx浓度明显降低,有GPx-3基因多态性的患者血浆GPx浓度下降更明显,神经功能缺损程度更重。Objective To study the change and its correlation between plasma concentration of glutathione peroxidase and polymorphisms of the glutathione peroxidase (GPx-3) promoter gene in patients with acute cerebral infarction(ACI). Methods 94 patients with ACI and 80 normal controls were detected the concentration of plasma glutathione peroxidas. The patients with ACI were undertaken neural function deficient scale(NDS) after 1 d onset. GPx-3 gene polymorphism were detected using polymerase chain reaction and ligase detection reaction(PCR-LDR) in patients with ACI. Results Plasma glutathione peroxidase concentration in ACI group were lower than that in the normal control group in first day onset ( P 〈 0. 01 ). There was not significantly different in the plasma glutathione peroxidase concentration between ACI group and normal control group in 14th day onset ( P 〉 0. 05 ). The plasma concentration of glutathione peroxidase and NDS in patients with mutation of GPx-3 promoter gene were obviously lower than those in the patients without mutation ( P 〈 0. 05 - 0.01 ). Conclusions The concentration of plasma glutathione peroxidase is decreased in ACI patients, and it is obviuosly lower in the patients with mutation of GPx-3 promoter gene, who are more seriously in neurological dysfunction.
