摘  要:目的:探讨抗抑郁药氟西汀对抑郁模型大鼠海马区胶质细胞源性神经营养因子(GDNF)mRNA表达的影响。方法:将雄性SD大鼠随机分为对照组、对照加药组、抑郁组及抑郁加药组4组。以慢性轻度不可预见性的应激结合孤养法建立抑郁动物模型,对照加药组、抑郁加药组予氟西汀5 mg.kg-1.d-1腹腔注射,对照组、抑郁组予蒸馏水腹腔注射。采用旷野试验和蔗糖水消耗实验观察大鼠行为改变,荧光实时定量PCR检测各组大鼠海马区的GDNF mRNA表达水平。结果:应激21 d后,抑郁组和抑郁加药组海马区的GDNF mRNA表达水平显著高于对照组和对照加药组(P<0.05和P<0.01)。结论:应激可能通过机体的反馈调节作用引起大鼠海马区GDNF mRNA的表达增高,氟西汀对大鼠海马区GDNF mRNA的表达无明显影响。Objective To explore the effect of antidepressant drug(fluoxetine) on the mRNA expression of glia cell line-derived neurotrophic factor(GDNF) in the rat hippocampus of depression model.Methods Male Sprague-Dawley rats were divided into control group,control+fluoxetine group,depression model group and depression model+fluoxetine group.The depression model of rat was established by stimulation of separation and chronic mild unpredicted stress.The behavior was measured by open-field and sucrose water consumption test.The mRNA expression of GDNF in the rat hippocampus was assayed by Real-Time Reverse-Transcription PCR.Results The mRNA expression of GDNF in the rat hippocampus was significantly different in the four groups on the 21st day after the beginning of the stress(F=7.47,P〈0.01),the mRNA expression of GDNF in the rat hippocampus of the depression model group and the depression model+fluoxetine group were significantly higher than that of the control group and the control+fluoxetine group(P〈0.01 or P〈0.05).Conclusion The stress may increase the mRNA expression of GDNF in the rat hippocampus in a feed-back regulation mode.The fluoxetine has no effect on the mRNA expression of GDNF in the rat hippocampus.
