摘  要:目的:观察罗格列酮对糖尿病大鼠钙化血管的治疗作用及对骨转录因子Msx2表达的影响。方法:在高脂饮食加小剂量链脲佐菌素腹腔注射处理的基础上,加用维生素D3和尼古丁处理,制备糖尿病合并血管钙化大鼠模型(糖尿病合并血管钙化组),给予部分糖尿病合并血管钙化组大鼠罗格列酮灌胃(2 mg.kg-1,每天1次,共8周)处理(罗格列酮组),同时设立空白对照组。实验第21周末,处死各实验组大鼠,检测各组大鼠的代谢指标,进行血管von Kossa染色,检测血管钙含量和碱性磷酸酶活性,应用real-time PCR方法检测Msx2 mRNA及蛋白免疫印迹方法检测Msx2蛋白在血管中的表达变化。结果:与对照组相比,糖尿病合并血管钙化组大鼠血管内存在弥漫性的钙盐沉积,血管内钙含量增高,碱性磷酸酶活性增强,Msx2 mRNA和蛋白表达明显增高(均P<0.05);而给予罗格列酮处理后,钙盐在血管内的沉积明显减少,钙含量和碱性磷酸酶活性分别下降8.37%、10.59%,Msx2 mRNA和蛋白表达也分别下降36.73%、42.55%(与糖尿病合并血管钙化组相比,均P<0.05)。结论:罗格列酮可以延缓糖尿病大鼠血管钙化的发展,并可以减少钙化血管中Msx2的表达。Objective To investigate the effect of rosiglitazone on aorta calcification of diabetic rats, and the ex- pression of Msx2 in calcified artery of diabetic rats. Methods Male Wistar rats were received a single low dose of strep- tozotocin (STZ) after 8 weeks of a high fat diet to induce diabetes, and then treated with vitamin D3 and nicotine ( DM + VDN group). Half of them were received rosiglitazone (2 mg" kg - 1 per day for 8 weeks, RSG group). Measurements of metabolic parameters ,yon Kossa calcium stain, aortic calcium content and alkaline phosphatase (ALP) activity, real-time PCR for Msx2 mRNA, and western blotting for Msx2 protein were performed. Results In the DM + VDN group, calcium deposition in aorta, aortic calcium content, ALP activity and expression of Msx2 mRNA and protein in calcified artery in- creased significantly as compared to control group (P 〈 O. 05 ). 8 weeks of rosiglitazone treatment decreased the calcium deposition in aorta, respectively decreasing aortic calcium content and ALP activity by 8.37 %, 10.59 %. The expres- sion of Msx2 mRNA and protein in calcified artery were also decreased by 36.73 % ,42.55 % compared to DM + VDN group (P 〈 0.05). Conclusions Long-term rosiglitazone treatment can attenuate the effect of diabetes on rat vascular calcification, and down-regulate the expression of Msx2 mRNA and protein in calcified artery.
