摘 要:目的观察重症急性胰腺炎(SAP)大鼠脑内脑红蛋白(Ngb)的表达水平变化。方法应用4%和0.5%的牛磺胆酸钠(sod ium taurocholate,STC)溶液诱导大鼠重症急性胰腺炎(SAP)和轻症急性胰腺炎(MAP)模型。同时设立假手术对照组。分别于模型成功诱发后4、8、16和24 h取脑组织标本,免疫组化方法检测Ngb阳性细胞在各组大鼠脑组织表达分布情况,用图像分析软件进行定量分析并作统计学比较。结果SAP和MAP模型均复制成功。模型诱发后8 h,SAP组脑内Ngb阳性细胞增多,其表达水平明显高于MAP组和假手术组(P<0.05);以后随时间延长而继续升高,24 h时Ngb表达显著升高(P<0.01)。结论SAP早期大鼠脑组织中Ngb表达明显增加,可能发挥对脑组织缺氧的适应性保护作用,同时提示缺血缺氧状态的持续存在是维持和促进胰性脑病病情发展的重要因素。Objective To investigate the intracephalic neuroglobin (Ngb) fluctuation in rats severe acute pancreatitis model. Methods Rats SAP and MAP models were incubated using sodium taurocholate (STC) solution of 4% and 0.5% respectively. Sham group was treated as protocol. Immunohistochemistry staining was used to investigate the distribution of Ngb positive ceils in rats brain of each group at 4 h, 8 h, 16 h and 24 h after model incubation. Quantitative image analysis software was used to perform statistical analysis. Results SAP and MAP models were duplicated successfully. After the SAP model incubation 8 hours, Ngb level in brain tissue was elevated compared with MAP and Sham group (P 〈0.05) and significantly increased at 24 h. Conclusion In early stage of SAP, Ngb level which increases significantly in brain may perform the neuroprotective effect in hypoxia condition. The persistence of ischemia and hy- poxia condition in brain may play an important role in aggravation of pancreatic encephalopathy.
