北方汉族人群TH01、TPOX、D6S1043和D12S391基因座遗传多态性研究

摘　　要：目的:对THO1,TPOX和D6S1043,D12S391基因座在北方汉族群体中的法医学应用价值进行比较。方法:用chelex100法提取445名北方汉族无关个体血痕的DNA,分别采用Identifiler和Sinofiler系统复合扩增,用ABI3130XL基因分析仪进行毛细管电泳,GeneMapper IDv3.2软件进行分型,计算四个基因座的多态信息。结果:四个基因座基因型分布符合Hardy-Weinberg平衡。统计结果计算显示D6S1043、D12S391、TH01和TPOX基因座的杂合度(H)分别为0.87、0.845、0.67和0.662:个人识别能力(DP)分别为0.968 6、0.951 0、0.856 2、0.805 6:多态信息含量(PIC)分别为0.861、0.816、0.633、0.576,非父排除率(PE)分别为0.682 4、0.635 0、0.382 4、0.373 3。结论:D6S1043和D12S391在北方汉族人群中具有良好的多态性,更适合应用于北方汉族人群的个体识别与亲权鉴定。Objective： The aim was to investigate the polymorphisms of THO 1, TPOX, D6S 1043 and D 12S391 loci in Northem Chinese Han population and evaluate their values in forensic testing. Methods： By using chelexl00 methods, Identifiler and Sinofiler multiplex amplification systems, capillary electrophoresis and GeneMapper v3.2, 4 STR loci were genotyped in 445 unrelated individuals from Han population in Northern China. Genetics parameters were calculated by statistical methods. Results： The results of test for Hardy-Weinberg equilibrium showed that the genotype distributions were correspondent with the expected. The heterozygote （H） of D6S 1043, D 12S391, TH01 and TPOX loci was 0.870, 0.845, 0.670 and 0.662 respectively. The discrimination power （DP） was 0.968 6, 0.951 0, 0.856 2 and 0.805 6 respectively. Polymorphism information content （PIC） was 0.861, 0.816, 0.633, and 0.576 respectively. The probability of exclusion （PE） was 0.682 4, 0.635 0, 0.382 4 and 0.373 3 respectively. Conclusion： It is suggested that the D6S1043 and D12S391 loci be good markers in forensic haemogenetics, and more suitable for Northern Chinese Han population in forensic testing.
