摘 要:目的观察X连锁凋亡抑制蛋白(X-linked inhibitor of apoptosis protein,XIAP)对耐苯妥英纳和卡马西平大鼠电生理指标和行为学的影响。方法建立慢性杏仁核点燃癫痫模型,筛选出耐药组和有效组,通过侧脑室注射全硫代XIAP反义寡核苷酸,对照组分别注射生理盐水。观察其对各组大鼠后放电阈值(after dischargethreshold,ADT)、后放电时程(after discharge duration,ADD)等电生理指标和行为学的影响。结果相对于对照组,耐药组大鼠在给予XIAP反义寡核苷酸后,ADT明显高于对照组(P<0.05);ADD时程明显缩短,Racine行为分级明显下降(P<0.05)。结论下调XIAP表达可以协助AEDs改善耐药大鼠的电生理活动,降低点燃后发作分级,提示XIAP参与了难治性癫痫的耐药。Objective To invesgate the effect of XIAP on electrobiological activity and seizure behavior in PHT- CBZ resistant rats. Methods The model of medically intractable epilepsy was established by kindling of amygdale. XIAP of antisense oligonueleotides (AS ODNs) was applied to PHT-CBZ resistant rats by lateral ventricle, and after discharge threshold( ADT), after discharge duration (ADD) and seizure activity was observed. Results Compared with the control group, the ADT was higher in PHT-CBZ resistant rats after XIAP of AS ODNs was applied ( P 〈 0.05 ) ; XIAP of AS ODNs could shorten the ADD, reduce the Racineg degree ( P 〈 0.05 ). Conclusion Down-regulation of XIAP could improve the electrobiological activity and seizure behavior in PHT-CBZ resistant rats. It indicated that XIAP was involved in muhidrug resistance in medically intractable epilepsy.
