摘  要:目的本实验研究清醒状态下大鼠急性大量灌胃乙醇后海马齿状回(dentate gyrus,DG)区细胞外液部分氨基酸类神经递质含量的变化。方法将成年Wistar大鼠(270±30g)12只随机分成对照组(n=6)和酒精组(n=6)。酒精组大鼠胃内灌入50%食用酒精,每次0.5ml,每10min灌1次,直到大鼠出现酒精中毒状态为止。对照组给予同等剂量的生理盐水。各组动物用脑内微量透析法及高效液相色谱法测定海马DG区细胞外液中的几种氨基酸类神经递质的含量,包括谷氨酸(glutamate,Glu)、天冬氨酸(aspartate,Asp)、谷氨酰胺(Glutamine,Gln)、甘氨酸(Glycine,Gly)、牛黄酸(Taurine,Tau)和丙氨酸(Alanine,Ala)。结果与对照组相比,急性酒精中毒时大鼠海马DG区细胞外液中的Asp、Glu和Gly的含量均明显增加(P<0.05),而Gln、Tau和Ala含量无明显变化(P>0.05);结论急性酒精中毒与海马DG区Asp、Glu和Gly含量增加有关。Objective To investigate the effect of acute alcoholism on amino acids levels in hippocampus DG of conscious rats. Methods Twelve old rats were randomized into control group( n =6)and alcohol group( n =6). Alcohol group rats were orally exposed to 50% ethanol 0.5ml per 10minute, until the rat to the alcoholics condition. Control group were orally exposed to the same dose of saline. The mierodialysis technique and high performance liquid chromatography was used to quantify Glutamate (Glu), Aspartate ( Asp), Glutamine ( Gin), Glyeine (Gly), Tanrine ( Tau ) and Alanine (Ala) in hippocampus DG. Results The glutamate,asptamate and Glycine in ethanol group were significantly increased than that in Control group (P 〈 0.05,respectively). Conclusions Acute alcoholism is accompanied with Glu, Asp and Gly release in hippocampus DG.
