摘  要:目的:探讨丹参酮ⅡA(TanshinoneⅡA,TanⅡA)对大鼠肺缺血/再灌注损伤的保护作用及机制。方法:将40只SD大鼠随机分为四组:对照组、模型(I/R)组、低剂量干预组、高剂量干预组,每组10只。模型组和干预组用血管夹夹闭左侧肺门30 min,去除血管夹恢复血流2 h。干预组于缺血前3 d给予不同剂量丹参酮ⅡA灌胃。对照组和模型组灌服等量生理盐水。测定各组肺组织的湿干比(W/D)、肺组织中髓过氧化物酶(MPO)活性、丙二醛(MDA)的含量。结果:模型组MPO、MDA、以及W/D较对照组显著增高。干预组各项指标均明显低于模型组,高剂量干预组各项指标均明显低于低剂量干预组。结论:TanⅡA对肺缺血/再灌注损伤大鼠有显著的保护作用,其机制可能与抗氧化自由基损伤、抑制中性粒细胞激活有关。Objective: To investigate protective effects of Tanshinone ⅡA (Tan Ⅱ A) on isehemia - reperfusion lung iniury in rats and the possible mechanisms. Methods:40 SD rats were randomly divided into 4 groups: control group, model group(I/R), low dose Tan Ⅱ A group and high dose Tan Ⅱ A group. Tan ⅡA were administered to rats in experimental groups 3 days before test, while physiological saline were administered to rats in control and model groups at the same time. Isehemia - reperfusion injury were indueed by ligation of left pulmonary artery. Ratio of wet to drying (W/D), malondialdehyde (MDA) and myeloperoxidase (MPO) were determined. Results: W/D, MDA and MPO in model group were significantly higher than those in control group. Compared with W/D, MDA and MPO in model group, those in experimental group were significantly lower. Conclusion : Tan Ⅱ A protects lung from isehemia - reperfusion iniury in rats. The mechanism is related to its antioxidative effect and the inhibition of neutrophil activation.
