摘  要:目的研究肿瘤坏死因子相关凋亡诱导配体(TRAIL)对喉鳞癌裸鼠动物模型的抑制作用,探讨其可能的作用机制。方法建立人喉鳞癌裸鼠移植瘤模型,应用TRAIL进行瘤内注射治疗,观察肿瘤生长情况,瘤体积变化,观察裸鼠的生存及死亡发生情况,并且通过光镜及透射电镜观察肿瘤微观结构发生的改变。结果体内注射TRAIL 100 ng/(mL.d),裸鼠肿瘤生长受到明显抑制,与对照组瘤体积相比差异具有统计学意义(P<0.01)。光镜,电镜可以观察到实验组肿瘤发生坏死和凋亡改变。结论TRAIL对喉鳞癌裸鼠动物模型的生长及发展存在着抑制作用。其作用可能是通过诱导肿瘤细胞凋亡来实现的。Objective To investigate the therapeutic effect of tumor necrosis factor related apoptosis-inducing ligand( TRAIL) on human laryngeal squamous carcinoma in nude mice, and to explore the possible mechanism. Methods Nude mouse model bearing laryngocarcinoma was established by using human laryngeal squamous carcinoma cell line (Hep-2), and the animals were given TRAIL 100 ng/( mL·d) or PBS for 14 consecutive days. The volumes of the subcutaneous tumor were measured. Uhramicrostructural changes of carcinoma after administration of TRAIL were observed under light and electron microscopes. Results In vivo, the differences were statistically significant in the tumor volume between the treated group and the control group( P 〈 0.01 ). The necrosis and apoptosis of the tumor cells were found under light and electron microscope in the treated group. Conclusion TRAIL can significantly inhibit the growth of laryngeal squamous carcinoma Hep-2 cell line in vitro and the growth of laryngocarcinoma in nude mice in vivo, the inhibitory effects are resulted from the apoptosis-inducing effect of TRAIL.
