摘 要:目的研究不同气腹介质对人结肠癌细胞株LS-174T在体外实验中增殖情况的影响,探讨腹腔镜应用于结直肠癌手术的安全性与可行性。方法MTT法测定细胞增殖数的情况。软琼脂克隆形成试验测定单个细胞增殖能力。免疫组织化学检测HIF-1α、VEGF的表达。结果CO2气腹作用后人结肠癌细胞LS-174T增殖速度高于对照组(P<0.05),CO2、He人工气腹作用后结肠癌细胞中HIF-1α和VEGF的阳性表达率高于对照组(P<0.01)。结论CO气腹可促进结肠癌细胞LS-174T的生长,He气腹相对抑制其生长。Objective To discuss the safety and feasibility of artificial pneumoperitoneum in the colon carcinoma operation through the investigation about the influence of different pneumoperitoneal medium on colon carcinoma LS-174T cell proliferation in vitro. Methods The cell proliferation condition was measured by MTT. Single cell proliferational capacity was tested by soft agar clone formation experiment. The expression of HIF-1α and VEGF were examined by immunohistochemistry. Results The growth speed and proliferating capacity of colon carcinoma cell LS-174T after CO2 pneumoperitoneum effect were higher than control group(P 〈 0. 05). Positive expression rates of HIF-1α and VEGF in colon carcinoma cell affected by CO2 and He artificial pneumoperitoneum were higher than control group ( P 〈 0.01 ). Conclusion CO2 pneumoperitoneum can facilitate the growth of colon carcinoma cell LS-174T, while He pneumoperitoneum can inhibit its growth relatively.
