摘 要:目的观察侧脑室微量注射L—Dopa(L—Dopa)对帕金森病(PD)模型大鼠的行为及多巴胺能神经元的数量的影响。方法应用6-羟基多巴胺(6-OHDA)制备偏侧损毁的PD模型大鼠,埋置金属套管,侧脑室微量注射不同浓度的L—Dopa1周,观察不同时段大鼠行为的变化及黑质多巴胺能神经元数量的变化。结果侧脑室微量注射超过1g/L L—Dopa可明显减少由阿扑吗啡(APO)诱发的PD模型大鼠向健侧的旋转次数(P〈0.01),其效果在2h左右达到高峰,且多巴胺能神经元的数量也明显增多(P〈0.01)。结论侧脑室微量注射适当浓度的L—Dopa可有效地改善PD模型大鼠的旋转行为并能增加多巴胺能神经元的数量。Objective To investigate the effects of intraventricular microinjection of L-dopa on behaviors and the number of dopaminergic neurons in rat Parkinson disease (PD) models. Methods The rat models of hemiparkinsonism were established with 6-hydroxydopamine (6-OHDA), stainless steel guide cannulas (0.7 mm) was implanted, and L-dopa with different concentrations was microinjected into the intracerebral ventricle of the rats. One week later, changes in behaviors and the number of dopaminergic neurons in substantia nigra were monitored at different time points. Results Microinjeection of 1 g/L L-dopa could significantly decrease the rotations induced by APO in rat PD models (P 〈 0.01 ), and the effect achieved summit in 2 h. The number of dopaminergic neurons in substantia nigra wasalso increased markedly (P 〈 0.01 ). Conclusion Intraventricular microinjection of L-dopa can relieve the rotation behaviors and increase the number of dopaminergic neurons in rat PD models.
