摘  要:目的探讨白藜芦醇对高脂饲养大鼠肾脏的保护作用及其机制。方法30只雄性Wistar大鼠随机分为正常对照组(A组)、高脂喂养组(B组)和白藜芦醇治疗组(C组)。喂养16周后,检测各组大鼠的血糖(BS)、三酰甘油(TG)、胆固醇(TC)、肌酐(Scr)、尿素氮(BUN)水平及24h尿蛋白定量;测定肾组织中丙二醛(MDA)含量、谷胱甘肽过氧化物酶(GSH-Px)和铜、锌-歧化酶(Cu、Zn-SOD)活性;同时取肾组织作PAS染色;以及对核因子(NF)-κB和单核细胞趋化蛋白质-1(MCP-1)的表达进行免疫组织化学检测。结果与B组比较,C组大鼠体内BS、TG、TC差异没有显著性;而肾组织MDA含量降低,Cu、Zn-SOD及GSH-PX活性升高;肾小球基质面积减小;NF-κB和MCP-1的表达降低。结论白藜芦醇可改善高脂饲养大鼠的肾脏功能,其机制可能与其抗氧化、抑制NF-κB活性、降低MCP-1的表达有关。[ Objective ] To study the renal protection of resveratrol in rats with high-fat diet and its mechanism. [ Methods ] Thirty Wistar rats were randomly divided into normal group (group A), high-fat diet group (group B) and resveratrol group (group C). After 16 weeks, blood sugar (BS), triglyeerides (TG), total cholesterol (TC), creatinine (Scr), blood urea nitrogen(BUN) and 24 h-urinary protein quantity(24 hours Upro) were examined in the corresponding rats, and the level of malondialdehyde (MDA), Cu, Zn-superoxide dismutase (Cu, Zn-SOD) and glutathione peroxidase (GSH-Px) in renal tissues were detected respectively. Additionally, renal tissues were carried out histology staining with PAS and immunohistology staining to analyze the expression of Nuelear Factor Kappa B (NF-κB) and monocyte ehemoattractant protein-1 (MCP-1). [ Results] No significant differences of blood glucose and blood lipid were found between rats with high-fat diet and resveratrol treatment groups. Compared with the high-fat diet groups, the levels of serum creatinine, blood urea nitrogen, urinary protein excretory rate, MDA, MCP-1 and NF-κB protein in resveratrol treated group decreased significantly, but the activity of renal Cu, Zn-SOD and GSH-Px increased signifieandy. Renal pathologic changes in resveratrol treated group were also improved. [ Conclusion] Resveratrol improved the biochemical parameters and renal pathologic changes in rats with high-fat diet, which is closely related to the antioxidization and the decreasing of NF-κB and MCP-1 activity in renal tissus.
