S100及S100A4蛋白在非小细胞肺癌中的表达及意义

摘　　要：目的研究非小细胞肺癌(NSCLC)组织中S100和S100A4蛋白的表达,探讨S100和S100A4在NSCLC侵袭和转移中的作用。方法应用免疫组化EnVision法检测91例非小细胞肺癌及33例NSCLC癌旁正常肺组织中S100和S100A4蛋白的表达水平。结果NSCLC组织中S100和S100A4的阳性表达率均高于癌旁正常肺组织(P<0.05);S100和S100A4的表达与患者年龄无关(P>0.05);S100的阳性表达与肿瘤大小有关(P<0.05),而S100A4的阳性表达率与肿瘤大小无关(P>0.05);有淋巴结转移者S100的阳性表达率高于无淋巴结转移者(P<0.05);生存时间在1a内患者的S100和S100A4阳性表达率高于存活1a以上者(P<0.05)。在鳞癌患者中,Ⅲ期患者的S100和S100A4的阳性表达率均高于Ⅰ期患者(P<0.05),有淋巴结转移者S100A4的阳性表达率高于无淋巴结转移者(P<0.05);在腺癌患者中,Ⅲ期患者的S100的阳性表达率高于Ⅰ期和Ⅱ期患者(P<0.05)。结论S100和S100A4可能与NSCLC的侵袭、转移和预后相关,可作为评估NSCLC的侵袭、转移和预后的参考指标之一。Objective To determine the expression of S100 and S100A4 in non-small cell lung cancer （NSCLC） specimens and try to find out the possible roles of S100 and S100A4 in the invasion and metastasis of NSCLC. Methods Immunohistochemical ENVISION method was used to detect the expression of S100 and S100A4 in 33 normal lung tissues near the NSCLC and 91 NSCLC tissues. Results The expression levels of S100 and S100A4 in the NSCLC tissues were significantly higher than those of the control groups （P〈0.05）.The expression of S100 and S100A4 were not related to age （P〉0.05）. The expression rate of S-100 was correlated to the tumor volume （P〈0.05）, but the expression of S100A4 were not related to tumor volume （P〉0.05）. The positive rate of S100 and S100A4 in Ⅲ stage squamous carcinoma patients were remarkably higher than Ⅰ stage patients （P〈0.05）, and in Ⅱ stage adenocarcinoma patients the positive rate of S100 were higher than Ⅰ stage patients（P〈0.05）.The positive rate of S100 were significantly higher in NSCLC patients with lymph node metastasis than those without lymphatic metastasis （P〈0.05）. In squamous carcinoma patients with lymph node metastasis the positive rate of S100A4 were significantly higher than those without lymphatic metastasis（P〈0.05）. In the squamous carcinoma patients survival for under 1 year had much higher S100 expression rate than those survival for above 1 year （P〈0.05）.Conelusion S100 and S100A4 may play important roles in NSCLC invasion and metastasis, their expression may act as a reference to evaluate metastasis and prognosis of NSCLC.
