摘 要:目的探讨鱼藤酮对大鼠原代培养中脑细胞血红素加氧酶-1的影响。方法不同浓度的鱼藤酮处理原代中脑细胞培养不同时间,逆转录-聚合酶链反应(RT-PCR)和Western blot检测血红素加氧酶-1表达的改变。结果鱼藤酮可致神经元发生退变;血红素加氧酶-1表达水平,在鱼藤酮处理24h,0.01、0.1μmol/L浓度时增加显著,1.0μmol/L浓度组与对照组相比,差异无统计学意义;而1.0μmol/L鱼藤酮处理6h,血红素加氧酶-1表达显著升高,12和24h时无变化。而RT-PCR检测鱼藤酮处理24h后的血红素加氧酶-1表达水平无显著改变。结论鱼藤酮引起血红素加氧酶-1表达升高,可能与细胞受损程度有关。Objective To study the effect of rotenone on the expressions of heme oxygenase-1. Methods Mid-brain cells were treated by different concentrations of rotenone for different times. RT-PCR and Western Blot were used to measure the expressions of HO- 1. Results The degenarations of neurons were apparently observed. The expressions of HO-1 of mid-brain cells treated by 0.1 and 0.01 μmol/L of rotenone for 24 h and 1.0 μmol/L for 6 h. There were no changes of HO-1 at transcription level after 1.0 μmol/L of rotenone treated for 24 h. Conclusion The increase of the expression level of HO-1 in Mid-brain cells induced by rotenone could be assoaiated with the degree of cell damage.
