摘 要:目的观察乙醇中毒后,雄性大鼠下丘脑促性腺激素释放激素(GnRH)神经元合成和释放激素功能及交配行为的变化,以此来探讨急性乙醇中毒后大鼠行为改变的可能机制。方法健康Wistar雄性大鼠40只,随机分为4组:对照组、低浓度乙醇组、中浓度乙醇组和高浓度乙醇组。实验组大鼠灌服不同浓度的乙醇,对照组大鼠灌服同等剂量的0.9%氯化钠溶液,2h后,观察以下指标:(1)交配行为;(2)血液乙醇浓度(BAC);(3)血清睾酮(T)水平;(4)下丘脑GnRH神经元密度和染色反应的变化。结果与对照组相比,各实验组大鼠BAC逐渐升高,骑跨次数和插入次数明显减少,差异有统计学意义(P<0.05),骑跨潜伏期及插入潜伏期显著延长(P<0.01);血睾酮水平较对照组均降低,高浓度乙醇组大鼠下丘脑内GnRH阳性神经元密度和染色强度较对照组明显减小(P<0.01),中、低浓度乙醇组下丘脑内GnRH阳性神经元密度和染色强度较对照组变化不显著(P>0.05)。结论急性乙醇中毒后,大鼠交配行为的改变可能与下丘脑GnRH神经元的合成和分泌功能被抑制,并引起血中睾酮水平相应降低有关。Objective To observe the changes in neuron synthesis of hypothalamic gonadotropin - releasing hormone (GnRH) and the releasing hormone functions and in mating behavior in male rats, thus to explore the possible mechanisms of behavior changes induced by acute alcoholism and its social significance. Methods Forty healthy male Wistar rats were randomized into groups control, low ethanol, medium, and high. Experimental groups were fed with different concentrations of ethanol, and the control with 0. 9% sodium chloride solution in the same dose. Two hours later, mating behaviors, blood alcohol concentration (BAG), serum testosterone (T) level and changes in hypothalamic GnRH neuronal density and staining reaction were observed. Results When compared with control, BAC increased gradually and the frequencies of mounting and intromission reduced significantly in experimental groups, the difference was significant ( P 〈 0.05 ), and the latent period was pro- longed significantly (P 〈 0. 01 ) ; serum testosterone level was lower in experimental groups than in control, hypothalamic GnRH positive neuronal density and staining intensity decreased significantly in high ethanol group (P 〈 0. 01 ), but not changed significantly in groups medium, low ethanol ( P 〉 0. 05 ). Conclusion After acute ethanol intoxication, changes in rat sexual behavior may be associated with inhibition of hypothalamic GnRH neuronal synthesis and secretion function and with decrease of serum testosterone level.
