摘 要:以鲫鱼(Carassius auratus)为试验材料,研究了在一定环境条件下重金属锌(Zn)的胁迫对鲫鱼不同组织中金属硫蛋白(MT)含量的影响。结果表明,在水温为(26.5±1.0)℃时,平均体长为(15.32±0.63)cm、平均体重为(310.6±5.7)g的鲫鱼不同组织中的MT本底值存在着明显的差异。鳃丝、肌肉、肝脏和肾脏中的MT含量范围分别为(1.024±0.018)^(1.081±0.019)μg/g、(1.171±0.021)^(1.205±0.015)μg/g、(2.589±0.013)^(2.706±0.015)μg/g和(2.262±0.009)^(2.453±0.003)μg/g,MT含量顺序为肝脏>肾脏>鳃丝>肌肉。在0.1 mg/L、0.5 mg/L、1.0 mg/L、10.0 mg/L Zn2+的胁迫下,鲫鱼的4种组织中MT含量的总体变化趋势较为一致,都是呈先升高后稳定的状态,在试验后6 h内的增加速率最大。其中鲫鱼的鳃丝、肝脏和肾脏3种组织中MT含量在24 h时达到峰值,随后略有下降并趋于稳定。而肌肉中的MT含量在24 h时没有出现峰值,其随着作用时间的延长逐渐达到峰值。鲫鱼的4种组织在24 h内MT的增加量与Zn2+的质量浓度呈较好的相关性,表现出一定的剂量-效应关系,表明水体中的Zn2+可诱导鲫鱼组织中MT的合成与表达,且诱导时间主要在24 h之内。以上结果表明,鲫鱼肝、肾、鳃丝等组织中的MT可作为评价外源重金属Zn污染的生物标志物。The impact of waterborne zinc on the content of metallothionein (MT) in different tissues of Carassius auratus in experimental condition was investigated. When the water temperature was (26. 5±1.0), average body length of the Carassius auratus was ( 15.32 ±0. 63 ) cm and the middleweight was ( 310.6±5.7 ) g, The MT background values was distinctly different in different Carassius auratus tissues. The concentrations of MT in gill, muscle, liver and kidney was ( 1. 024±0. 018) - ( 1. 081±0. 019) μg/g, ( 1. 171±0. 021 ) - ( 1. 205±0. 015) μg/g, (2. 589±0. 013) - (2. 706±0.015) μg/g and (2.262±0.009) -(2.453±0.003) μg/g, respectively. The order of MT concertration was liver 〉 kidney 〉 gill 〉 muscle, and this order was comparatively steady. Induced by Zn^2+ (0. 1 mg/L,0. 5 mg/L, 1.0 mg/L and 10. 0 mg/L) , the MT contents in the four organs generally showed the same changing tendency: increased gradually at first and then changed steady and the increase rate reached a maximum in 6 h. The MT contents in gill, liver and kidney got up to maximum in 24 h, then decreased a little and kept steady. However in muscle, MT content didn't peak in 24 h but was up to max as exposure time prolonged. The increase of MT contents had a positive relation with the concentration of Zn^2+ in water, and showed dose-response relationship. It indicated that Zn^2+ in water can induce MT systemization and its expression in Carassius auratus tissues and the inducement maily appeared in 24 h. The research indicated that the content of MT in liver, kidney and gill can be used as a biomarker to evaluate the pollution of exogenous heavy metal zinc.
