摘 要:[目的]分析并明确中国汉族人群脂联素(adiponectin,AMP1)基因第二外显子45位T/G多态性(T45G)与2型糖尿病(type2 diabetes mellitus,T2DM)的相关性。[方法]对以往有关T45G与T2DM相关性研究进行Meta分析。通过文献检索收集中国汉族人群AMP1基因SNP45多态性与T2DM的病例-对照研究,剔除不符合要求的文献,对入选文献进行异质性检验并根据检验结果进行数据合并,采用Stata10.0计算合并比值比(oddsratio,OR)及其95%可信区间(confidence interval,CI)以及发表偏倚。[结果]共16篇文献符合条件纳入研究,采用随机效应模型合并OR值,Meta分析结果为在显性、隐性、加性遗传模式下,合并OR(95%CI)分别为1.61(1.21~2.14)、2.16(1.59~2.92)、1.49(1.22~1.83),OR值具有统计学意义。[结论]不论是显性、隐性还是加性遗传模式,G等位基因均为T2DM的危险因素,提示G等位基因携带者易患T2DM。[Objective]To identify the association between T/G polymorphism of adiponectin (AMP1) gene which located at 45 locus in exon 2 and type 2 diabetes mellitus (T2DM) in Chinese Han ethnic group. [Methods] Meta-analysis was carried out after all relevant case-control studies were collected. Odds ratios (OR) of AMP1 genotype to T2DM patients were calculated. Heterogeneity among individual studies, summary ORs and corresponding 95% confidence interval (CI), publication bias were estimated in Stata 10.0. [Results] Sixteen studies with 2 559 cases and 2 095 controls were collected. The geno- type frequencies were consistent with Hardy-Weinberg equilibrium. The frequencies of TT, TG and GG genotypes were 44.94%, 43.57% and 11.49% in case group and 55.37%, 38.52% and 6.11% in control group, respectively. The frequencies of allele G were 33.27% in cases and 25.51% in controls. The summary ORs (with 95% CI) under dominant model, recessive and additive models were 1.61 (1.21-2.14), 2.16 (1.59-2.92) and 1.49 (1.22-1.83), respectively. [Conclusion] In Chinese Han population, the allele G is a risk factor to T2DM under any heredity models, which suggested that G allele carriers are susceptible to T2DM.
