摘 要:建立了高效阴离子交换色谱积分脉冲安培检测器同时分离并测定注射液中18种常见氨基酸、氨基己酸和牛磺酸含量的方法。注射液用20mg/L的NaN3溶液稀释1000倍,经0.22um尼龙膜过滤后直接进样分析。以一定浓度的NaOH和NaAc溶液为淋洗液,选择合适的梯度淋洗条件,20种氨基酸在AminoPacPA10阴离子交换色谱柱上在30min内很好地分离,并用脉冲安培检测器进行了测定。氨基酸的检出限在0.14~3.81pmol(25uL进样,峰面积定量)。In the present paper, twenty amino acids, including 18 common amino acids, ε-aminocaproic acid and tanfine, have been separated and detected by high-performance anion-exchange chromatography with integrated pulsed amperometric detector. Injections were diluted 1000-fold with 20 mg/L NaN3 , and then filtered with 0.22 tan nylon membrane before injection and separation with NaOH and NaAc gradient elution. Twenty amion acids were separated in 30 min on anion-exchange column and then detected by integrated pulsed amperometric detector. The detection limits range from 0.14 to 3.81 pmol (25 uL injection, calculated as peak area). The proposed method has the advantages of simple operation, high sensitivity, good reproducibility, no interference and no need for derivatization.
