摘 要:目的探讨C-met、MAGE-1在原发性肝细胞癌中的表达及临床意义。方法采用免疫组织化学方法检测40例肝癌组织中C-met、MAGE-1的表达情况,并研究肝癌细胞中C-met、MAGE-1的表达及其与临床病理指标、侵袭转移之间的关系。结果C-met表达与肿瘤数目(χ2=7.697,P<0.05)及Edmondson分级(χ2=8.209,P<0.05)有关。MAGE-1表达与Edmondson分级有关(χ2=6.030,P<0.05)。C-met阳性表达患者术后1年复发率较阴性表达患者无明显差异(χ2=0.879,P>0.05),而术后2年复发率明显较高(χ2=12.485,P<0.05)。MAGE-1阳性表达患者术后1年复发率较阴性表达患者明显升高(χ2=5.393,P<0.05),而术后2年复发率无明显差异(χ2=0.287,P>0.05)。有C-met和(或)MAGE-1阳性表达的患者术后1年及术后2年复发率均较阴性表达患者明显升高(χ2=5.522,P<0.05,χ2=8.152,P<0.05)。C-met、MAGE-1在肝细胞癌组织中阳性率分别为65.0%、72.5%,在正常肝组织中分别为10.0%、0%,两组比较均有显著性差异(P<0.05)。C-met联合MAGE-1检测阳性率为85.0%。结论①C-met的表达与肝癌的肝内复发转移有关,是肝癌早期复发转移的风险因素。②MAGE-1在PHC患者肝癌组织中特异性高表达,可用来监测癌细胞扩散情况。③C-met、MAGE-1联合检测可提高诊断肝癌侵袭性的灵敏度。Objective To study the expression of C-met, MAGE-1 in hepatoceUular carcinoma and clinical significance. Methods Immunohistochemistry technique was used to evaluate the expression of C-met and MAGE-1, and the relationship between their expression and the clinic pathological prognostic parameters, invasion, metastasis was analyzed in 40 PHC. Results The positive rate of C-met was correlated with tumor number and Edmondson grade. While the expression of MAGE-1 was correlated with Edmondson grade. The positive rates of C-met in patients with recurrence in 1 year were higher than that in patients without recurrence, but no difference in 2 year. The positive rates of MAGE-1 in patients with recurrence in 2 year were higher than that in patients without recurrence, but no difference in 1 year. The positive rates of C-met and MAGE-1 with recurrence were higher than that in patients without recurrence in 1 year and in 2 year. Expression of C-met and MAGE-1 in PHC was 65.0%, 72.5%, respectively, but in normal liver tissue was 10.0%, 0%,respectively. The Combine expression of C-met and MAGE-1 was 85.0%. Conclusion (1)The expression of C-met positively related the tmnor recurence. (2)The higher frequency of MAGE-1 gene expression in PHC suggests the possibility of MAGE-1 gene encoding protein as target for monitoring the diffusion of tumor cells. (3) The unite detection of C-met and MAGE-1 could increase the sensitivity to diagnosis the invasion of hepatoma.
