摘  要:目的:观察核转录因子-κB圈套寡核苷酸抑制NF-κB活性后,肝癌HepG2细胞对ciglitazone敏感性的变化。方法:运用基因转染技术将NF-κB圈套寡核苷酸转染入肝癌HepG2细胞中,检测转染前后NF-κB活性。再用ciglitazone处理转染细胞,描记HepG2细胞生长曲线,流式细胞仪检测细胞周期,Western blot检测处理前后细胞周期素D1(Cyclin D1)的变化。结果:转染后的肝癌HepG2细胞NF-κB活性明显降低;ciglitazone抑制HepG2细胞增殖作用增强,70.65%的细胞被阻滞于G1/G0期,Cyclin D1表达明显下降。结论:NF-κB圈套寡核苷酸能增加肝癌HepG2细胞对ciglitazone的敏感性,可能与其下调NF-κB的活性、增强ciglitazone抑制肝癌细胞增殖和阻滞细胞周期有关。Objective To investigate the sensitivity changes of hepatoma cell to ciglitazone after the activity of NF-κB was inhibited with NF-κB decoy oligodeoxynucleotides(ODNs). Methods NF-κB decoy ODNs were transfected into HepG2 cells with lipofectamine, the activity of NF-κB was determined before and after transfection, respectively. The transfected HepG2 cell was treated with eiglitazone and then the growth was recorded with growth curve, the cell cycle was detected with by flow cytometry,the level change of Cyclin D1 was determined with Western blot. Results The activity of NF-κB in transfected HepG2 cells was significantly decreased; the inhibitory effect of ciglitazone on the proliferation of HepG2 cell was enhanced, 70.65% ceils was arrested in G1/G0 phase,the expression of Cyclin D1 was obviously decreased. Conclusion NF-κB decoy ODNs could improve the sensitivity of HepG2 ceils to ciglitazone, and the effect may relate with the downregulation of NF- κB activity,enhancing the inhibitory effect on the proliferation of hepatoma cells and arresting the cell cycle.
