摘 要:目的采用大鼠大脑中动脉栓塞(Middle cerebral artery occlusion,MCAO)模型及CT灌注成像(CT perfusion imaging,CTPI),结合TTC(triphenylte-trazolium chloride,TTC)染色,研究CTPI在脑缺血模型中的应用价值。方法将30只SD大鼠随机分为假手术组(A组)、缺血2h组(B组)和缺血6h组(C组),每组各10只。各组动物于不同时间点行CTPI检查。扫描完成后立即处死大鼠行TTC染色。结果A组动物灌注图两侧对称,无灌注异常区。B组和C组灌注图可见右侧大脑中动脉供血区明显的异常灌注。TTC染色显示A组两侧大脑半球呈均匀一致红染,B组见右侧纹状体及额顶叶皮质失染呈苍白色,C组失染区范围大于B组,而且边缘清晰。结论CTPI及MCAO模型适合于研究大鼠的脑血流动力学变化,可以反映脑缺血的范围。Objective To investigate the application value of CT perfusion imaging in middle cerebral artery occlusion model in rats and compared with TTC staining. Methods All 30 rats were divided into 3 groups: Group A (sham-operated for control study ); Group B (occluded 2 hours); Group C (occluded 6 hours). All rats were killed immediately after CTPI scan and dealt with TTC staining. Results In group A (sham-operated for control study), no abnormal outcomes were found in CTPI and TTC staining. In group B and C, CTPI showed abnormal perfusion in region of middle cerebral artery blood-supply. In group B, the region of lose staining were seen in the right striatum corpora and cortex of sinciput and parietal lobe with TTC staining. The region of lose staining in group C was larger than in group B. Conclusion The CT Perfusion Imaging and the middle cerebral artery occlusion model in rats can reflect the haemodynamics changes of cerebral ischemia.
