摘 要:目的探讨子宫内膜癌组织中多药耐药基因(MDR1)和多药耐药相关蛋白(MRP)基因的表达及其意义。②方法利用半定量逆转录聚合酶链反应(RT-PCR),对37例初治子宫内膜癌及10例正常子宫内膜组织MDR1和MRP基因的表达进行检测。③结果37例子宫内膜癌和10例正常子宫内膜组织均有MDR1和MRP基因的表达,两种基因在这两种组织中的表达水平无显著性差异(t=1.17,1.51,P>0.05)。有7例子宫内膜癌MRP基因呈高水平表达(1.68±0.17),明显高于其他30例病人(0.83±0.11),差异有极显著性(t=16.55,P<0.001)。④结论子宫内膜癌对化疗的耐药可能是由于固有性多药耐药所致,与MDR1和MRP基因的表达有关。Objective To study the expression and implication of multidrug resistance gene(MDR1) and multidrug resistance associated protein (MRP) gene in endometrial carcinoma. Methods Using semiquantitative reverse transcription PCR(RT-PCR), the expression level of both MDR1 and MRP genes were detected in endometrial carcinoma from 37 primary untreated patients (1995.7—1997.10) and 10 physiological endometriums. Results The expression of both MDR1 and MRP genes were detected in all endometrial carcinoma samples, as well as in all normal endometrium,the differences of the expression level of both MDR1 and MRP genes between the endometrial and normal endometrium were not significant(t=1.17,1.51,P>0.05).High expression of MRP gene(1.68±0.17) was shown in 7 endometrial carcinoma samples,which was much higher than that of other 30(0.83±0.11) ones (t=16.55,P<0.001). Conclusion The multidrug resistance(MDR) in endometrial carcinoma may be an intrinsic one, and it may be related to the expression of MDR1 and MRP genes.The overexpression of MRP gene may be as a prognostic factor for endometrial carcinoma patients.
