摘 要:目的探讨4种常见实体瘤多药耐药性与P-糖蛋白(P-gp)及p53表达的关系。②方法采用MTT比色法检测胃癌、大肠癌、乳癌、肺癌对14种化疗药的耐药性;用免疫组化方法检测同一标本P-gp,p53的表达。③结果109例标本中多药耐药者51例,疏水性药物耐药者59例。各肿瘤P-gp阳性表达者共40例,p53阳性表达者共63例。乳癌疏水性耐药组P-gp表达率为57.1%,对照组表达率为9.0%;大肠癌多药物耐药组p53表达率为100%,敏感组表达率为50%.④结论P-gp阳性表达可反映乳癌的疏水性耐药,p53阳性表达是大肠癌多药耐药的标志之一。Objective To investigate the relationship between the multidrug resistance and the expression of p-glycoprotein(P-gp) and p53 protein in 4 commonly seen solid tumors.MethodsSensitivities to 14 anticancer drugs were examined in gastric cancer, colorectal cancer, lung cancer and breast cancer with MTT assay and the expression of Pgp and p53 was detected by using immunohistochemistry.ResultsOf the 109 cases, 51 exhibited pleiotropic drug resistance and 59 cases had hydrophobic drug resistance,40 patients showed positive expression of P-gp and 63 patients were p53 positive.The P-gp positive rate of breast cancer patients with hydrophobic drug resistance was 57.1% and the control group was 9.0%.The expression rate of p53 in pleiotropic chemoresistance group of colorectal cancers was 100% and the control group was 50%.ConclusionThe hydrophobic drug resistance of breast cancer was associated with P-gp expression. p53 expression was the marker of pleiotropic drug resistance of colorectal cancer.
