摘  要:目的:在观察艾灸对实验性RA大鼠抗炎效应的基础上,研究艾灸对海马、下丘脑及垂体GR表达的影响,探索艾灸治疗RA的GR机制。方法:将雄性SD大鼠随机分为空白对照组、模型组、艾灸组,用福氏完全佐剂塑造大鼠实验性RA病理模型,艾灸"肾俞"、"足三里"治疗,测定大鼠右后足容积,采用免疫组化法检测海马、下丘脑、垂体GR表达。结果:造模后模型组大鼠足肿胀度明显高于空白对照组(P<0.01),艾灸组大鼠足肿胀度明显低于模型组(P<0.01);海马GR表达,模型组与空白组比较有增高趋势(P>0.05),艾灸组明显低于模型组(P<0.01);下丘脑GR表达,模型组明显高于空白对照组(P<0.01),艾灸组较模型组明显降低(P<0.01);垂体GR表达,模型组较空白对照组显著升高(P<0.01),而艾灸组与模型组比较无显著性差异(P>0.05)。结论:艾灸对实验性RA具有抗炎效应,艾灸对RA大鼠海马、下丘脑GR表达的调控可能是其实现抗炎效应的重要机制。Objective: On the basis of observing the moxibustion for experimental rheumatoid arthritis(RA) model rats′ anti-inflammatory effect,to study the effect of glucocorticoid receptor(GR) expression in hippocampus,hypothalamus and pituitary by moxibustion,to explore the mechanism of moxibustion to treat RA.Methods: Male Sprague-Dawley(SD) rats were randomly divided into control group,model group,moxibustion group,To produce RA animal models by Freund′s Complete Adjuvant(FCA),to moxibust in "Shenshu(BL23)" and " Zusanli (ST36)" acupoints. To detect their right feet capacity, and to detect GR expression in the hippocampus, hypothalamus, pituitary by immunohistochemistry method. Results : Model group rats' feet swelling were significantly higher than control group ( P 〈 0.01 ), moxibustion group rats' feet swelling were significantly lower than model group (P 〈 0.01 ). About GR expression in hippocampus, model group had the markup tendency compared with control group( P 〉 0.05 ), but moxibustion group decreased significantly than model group (P 〈 0.01 ). About GR expression in hypothalamus, model group was higher than control group (P 〈 0.01 ), but moxibustion group decreased significantly than model group (P 〈 0.01 ). About GR expression in pituitary, model group was higher than control group( P 〈 0.01 ), but there were no significant difference in moxibustion group and model group (P 〉 0.05 ). Conclusion: Moxibustion for experimental RA model rats' has anti - inflammatory swelling. It's maybe the important mechanism that moxibustion effectively regulates and controls hippocampus, hypothalamus GR expression to achieve anti - inflammatory effect.
