逐痰通络汤对大鼠局灶性脑缺血损伤的保护作用

摘　　要：目的:观察逐痰通络汤对大鼠局灶性脑缺血损伤的保护作用。方法:清洁级雄性SD大鼠18只,随机分为假手术组(K组)、模型组(C组)和逐痰通络汤组(Z组),应用线栓法建立大脑中动脉栓塞(MCAO)模型。造模后第1、3、7天观察各组动物神经功能缺损并评分,脑组织TTC染色并测定梗死体积,光镜观察脑组织病理学改变。结果:C、Z组各时间点均出现不同程度神经功能缺损,Z组在第7天分值低于C组(P<0.05)。C、Z组各时间点均出现不同程度梗死,Z组在第7天梗死体积小于C组(P<0.05)。C、Z组左侧大脑皮质及皮质下区出现梗死灶,缺血区及周边存在萎缩变性的神经元且淡染,胞浆及血管周围空化,梗死灶周围炎性细胞浸润,Z组与C组相比,第7天上述表现较轻。结论:逐痰通络汤能够促进局灶性脑缺血大鼠神经功能缺损的恢复,减小梗死体积,减轻脑组织病理损伤,在缺血早期发挥保护作用。Objective ： To observe protective effect of Zhutan Tongluo Tang on focal cerebral ischemia in rat. Methods ： 18 Sprague -Dawley male rats were divided randomly into 3 groups： sham operation group （K） , model group （C） and Zhutan Tongluo Tang group （Z）. The middle cerebral artery occlusion （MCAO） model was established by the thread - ty- ing method. We measured the scores of neural functional defect; The tissues of brain were stained with TTC and measured infarct area on 1,3 and 7 days after operation. Results ： Rats of C and D group had neural functional defect with different degree. The scores of Z group showed lower than C group on the day 7 （ P 〈 0.05 ）. Rats of C and D groups had in- farct area with different degree. The volumes of Z group showed lower than C group on the day 7 （ P 〈 0.05 ）. C and Z groups showed an ischemic region in cerebral cortex and subeortex. There were many atrophied and degenerative neurons with lightly staining in isehemie region. Cavitation around endochylema and blood vessel, inflammatory cells infiltrated ischemic region. Z group was less significant change than C group on the day 7. Conclusion： Zhutan Tongluo Tang could promote the neural functional recovery of focal cerebral isehemie rat, decreased infarct volumes and lightened pathological damage of brain tissue.
