摘  要:目的探讨去整合素kistrin对人晶状体上皮细胞株SRA01/04的细胞凋亡的影响。方法取对数生长期的人晶状体上皮细胞株SRA01/04用于实验。分别用含有不同浓度(80μg.L-1、160μg.L-1、320μg.L-1)kistrin的完全培养基孵育8h、16h、32h,然后用流式细胞仪检测细胞凋亡情况,并与阴性对照组(kistrin浓度为0μg.L-1)比较。用含kistrin的终浓度为160μg.L-1的培养基孵育细胞,于24h、32h后进行吖啶橙染色,倒置相差荧光显微镜观察细胞凋亡形态学变化,并与对照组(kistrin浓度为0μg.L-1)比较。结果kistrin作用8h时,阴性对照组与80μg.L-1、160μg.L-1、320μg.L-1kistrin孵育细胞组细胞凋亡率分别为(1.29±0.43)%、(5.48±0.66)%、(11.23±1.16)%、(20.77±1.46)%,kistrin作用16h时不同组别细胞凋亡率分别为(3.04±0.12)%、(7.68±0.23)%、(19.76±2.22)%、(32.36±3.29)%,kistrin作用32h时不同组别细胞凋亡率分别为(4.75±0.08)%、(72.15±7.04)%、(54.09±3.77)%、(43.27±3.32)%;在每个时间点各浓度的细胞凋亡率与阴性对照组相比,差异均有统计学意义(P均<0.01)。与对照组相比,浓度为160μg.L-1的kistrin作用24h后,细胞形态可见改变,kistrin作用32h后细胞出现明显的各种凋亡形态学改变。结论去整合素kistrin对人晶状体上皮细胞株SRA01/04有诱导凋亡的作用,为药物防治后发性白内障提供了新思路。Objective To investigate the effects of disintegrin kistrin on cell apoptosis in human lens epithelial cell line SRA01/04. Methods Human lens epithelial cell line SRA01/04 at logarithmic growth phase was used in the study, and incubated with medium containing different concentrations of kistrin (80 μg ·L^-1 , 160 μg ·L^-1, 320μg ·L^-1 ) for 8 hours, 16 hours and 32 hours,respectively. Cell apoptosis was detected by flow cytometer, and compared with that of control group (the concentration of kistrin was 0 μg ·L^-1 ). Medium containing 160 μg ·L^-1 kistrin was were used to incubate cells. Acridine orange staining was taken at 24 hours and 32 hours after incubation, morphologic changes of cell apoptosis were observed with inverted phase contrast microscope, and they were compared with control group ( the concentration of kistrin was 0 μg ·L^-1 ). Results At control group and different concentrations of kistrin (80 μg ·L^-1 ,150 μg ·L^-1 ,320 μg ·L^-1) ,the cell apoptotic rates were (1.29 ±0.43)% , (5.48± 0.66)% ,(11.23 ±1.16)% ,(20.77 ±1.46)% at 8 hours after treatment, (3.04 ±0.12)%, (7.68 ± 0.23 ) % , ( 19.76 ± 2.22) % , ( 32.36 ± 3.29) % at 16 hours after treatment, ( 4.75 ± 0.08)% ,(72.15+7.04)% ,(54.09±3.77)%,(43.27±3.32)% at 32 hours after treatment. There was significant difference in apoptotic rate for cach concentration at every time point compared to that of control group( all P 〈 0.01 ). There were significant morphologic changes when the cells were incubated with 160 μg ·L^-1 kistrin for 24 hours, and obviously apoptotic morphologie changes were observed after treated with ldstrin for 32 hours compared with control group. Conclusion Disintegrin kistrin has the ability of inducing apoptosis for human lens epithelial cell line SRA01/04 ,which provides a new way for prevention of after-cataract with drugs.
