摘 要:为比较微量法和分光光度法的差异,以Trolox为参比物,PG、BHA和BHT为阳性对照品,比较其IC50和TEAC;以儿茶素的抗氧化活性为研究对象,对2种方法的相关性做Paired-samplettest。发现2种方法所得到的TEAC基本一致,4个标准品和儿茶素的抗氧化活性顺序一致,即:PG>儿茶素>BHA>Trolox>BHT;2种方法的相关系数r=0.998,显著性P=0.501。研究结果表明,分光光度法操作繁琐费时,用样量大;微量法操作简单快速,用样量小,准确性好,适合对微量天然产物抗氧化活性的评价。To compare microplate assay (MPA) with spectro-photometric method ( SPM ), Trolox as reference substance, PG, BHA and BHT as positive control, was assayed by two methods. IC50 and TEAC of antioxidant activity were compared and studied. The paired-sample t test of antioxidant activity of catechin was assayed. The results showed that TEAC from two methods was almost approximate, and the antioxidant activity was also same in two methods : PG 〉 catechin 〉 BHA 〉 Trolox 〉 BHT. The coefficient correlations of the paired samples was 0. 998 ,the significant value of the paired samples was 0.501. The resuhs showed that the SPM was tedious,time-consuming, much sample, and the MPA was simple, rapid, accurate,little sample for the screening antioxidant activity of natural products.
