粟酒裂殖酵母tRNA前体3′末端加工酶Trz1p功能的研究

摘　　要：[目的]研究粟酒裂殖酵母tRNA前体3′末端加工酶Trz1p在体内的功能,为以后进一步探索Trz1p的功能奠定基础。[方法]构建Trz1+的表达载体pREP82x-trz1-5 flag,通过醋酸锂转化法将pREP82x-trz1-5 flag转入yYH1菌株和Trz1+的缺失菌株ySS0319中,并进行随机孢子分析。[结果]Trz1p C-端添加5FLAG仍能促进tRNA前体3′末端的加工成熟,但Trz1-5flag不能救活Trz1+的缺失菌株。[结论]粟酒裂殖酵母Trz1p具有tRNA前体3′末端加工功能。[ Objective ] The aim was to research the function of the pre-tRNA 3＇-end processing pnzyme Trzl p in Schizosaccharomyces pombe, and lay a foundation for further exploring the function of Trzl p. [ Method] Constructing the expression plasmid pREP82x-trzl-Sflag of Trz1 , the plasmid was transformed into yYH1 strain and ySS0319 by Lithium Acetate transformation method,then carried out random spore analysis. [ Result] Trzlp-5FLAG can still promote pre-tRNA 3＇-end maturation,but Trzl-Sflag cannot rescue Trzl ＋ disruption. [ Conclusion] Trzlp of Schizosaccharomyces pombe has pre-tRNA 3 ＇-end processing function.
