摘 要:目的:探讨消减免疫法在分析肝癌细胞凋亡反应中瞬时表达的某些相关抗原分子的作用。方法:利用消减免疫法制备抗肝癌凋亡细胞相关抗原的抗体;用ELISA方法检测抗原、抗体反应。结果:应用消减免疫法制备的抗凋亡肝癌细胞相关抗原的多克隆抗体与肝癌凋亡细胞相关抗原的反应较强;与非凋亡肝癌细胞的抗原反应较弱。其抗原表位主要是蛋白质或多肽。结论:消减免疫法可用于分析肝癌细胞凋亡反应中瞬时表达的相关抗原分子;凋亡过程中有新的蛋白质表达。Aim: To explore the role of subtractive immunization techniques in analysis of some transiently expressed antigens of hepatoma apoptotic cells during apoptotic response. Methods: subtractive immunization techniques were used to prepare polyclonal antibodies for hepatoma apoptotic cells associated antigens. ELISA and immunoblot assay were used to detect the reaction of antibodies with antigens. Results: The antibodies reacted more strongly with hepatoma apoptotic cell associated antigens(HACAA) than with hepatoma cell antigens. The epitopes were proteins or peptides. Conclusion: Subtractive immunization techniques could be used to analyze the transiently expressed antigens of hepatoma apoptotic cells. Some new proteins expressed during apoptotic response could be selected by this antibody.
