摘  要:人肺癌细胞株 SPC-A1加入全反式维甲酸(ATRA)使其终浓度为10μmol/L。培养7d 后处理组和对照组分别进行 MTT、光镜、电镜及流式细胞仪检测发现处理组细胞增殖减慢,细胞聚集成片,胞浆中空泡增多,核浆比例下降,无明显染色体固缩及凋亡小体。细胞周期动力学分析发现处理后的细胞生长周期向 G_(0/1)期聚集,S 期和 G_2+M 下降。提示 ATRA 对体外培养的人肺癌 SPC-A1株有一定的生长抑制和诱导分化作用,但凋亡作用不明显。We added retinoic acid into the study group of the human lung cancer cell line SPC-Al and made the terminal concentration 10μmol/L.After 7 days of culture,the study group and control group were measured with MTT method,which showed that cell proliferation slowed down the study group comparing with the control group.With the light and electron microscopy,it was found that the cells in the study group tended to aggregate,the vacuoles inside the cytoplasm increased,no marked chromosome fragmentation and apoptosis body.Analysis of cell cycle showed in the study group the percentage of G0/1 increase while that of S and G2+M decrease.It was concluded that retinoic acid has a certain effect on inhibition and induction of differentiation over SPC-Al line in vitro,but no marked apoptosis.The exact mechanism needs further study.
