姜黄素Ⅲ与丝裂霉素联用抑制肝癌细胞BEL-7402的实验研究

摘　　要：目的研究姜黄素Ⅲ联合丝裂霉素抑制体外人肝癌细胞BEL-7402生长增殖的效果。方法体外培养人肝癌细胞BEL-7402,以四甲基偶氮唑蓝(MTT)法测2种药物对癌细胞的抑制率及联合用药时的抑制率。结果姜黄素Ⅲ、丝裂霉素均能抑制体外肝癌细胞增殖并呈时间与剂量依赖性。姜黄素10、20、40、80μmol/L与丝裂霉素1、2μmol/L联合用药24、48、72h的抑制率显著高于单独用丝裂霉素、姜黄素Ⅲ(P<0.05),两者呈现出相加的抗癌效果。结论姜黄素Ⅲ能有效抑制体外人肝癌BEL-7402细胞增殖,联合丝裂霉素用药时能提高丝裂霉素对人肝癌BEL-7402细胞的敏感性。Objective To investigate the effects of curcumin Ill combined with mitomycin on the proliferation of human hepatoma cancer cell-BEL-7402 in vitro. Methods The human hepatoma cancer cells-BEL-7402 were cultured in vitro, and the respective and combined suppressive effects of cucumin Ⅲ and mitomycin on the BEL-7402 cell proliferation were observed by MTT method. The data were analyzed by SPSS software. Results The only curcumin Ⅲ or mitomycin could obviously suppress the proliferation of cancer cell-BEL-7402 in vitro in the time and dose dependent manners. As compared with simple curcumin Ⅲ or mitomycin , combined curcumin Ⅲ at 10,20,40,80μmol/L with mitomycin at 1 μmol/L,2 μmol/L respectively increased significantly the inhibition rate on the growth of BEL-7402 cells （ P 〈 0.05 ）, which showed that there was a synergistic actions of the two drugs. Conclusion Curcumin Ⅲ can effectively suppress the growth of cancer cell-BEL-7402 in vitro, and combined mitomycin with Curcumin Ⅲ can increase the sensitivity of BEL-7402 ceils to mitomycin.
